The development of the immune system in the mouse involves the concerted action of antigen recognition and signaling pathways which ultimately determine the cell-specific activation of the immune response. A number of these pathways and their functional role in determining the outcome of the immune response are under investigation. The signaling pathways of thymocyte maturation and T cell receptor (TCR) triggering will be explored. We have shown that TCR triggering in the thymus is under the regulation of the Notch signaling pathway. We will examine the requirements for Notch signaling in thymocyte development and TCR-mediated signaling. Since the role of the Notch signaling pathway in T cell development is novel, studies are proposed to identify the ligand for Notch in thymocyte development. Also under investigation is the role of a novel family of immunoglobulin superfamily proteins, the B7 family, in the regulation of the T cell immune response. The regulation of T cell activation and tolerance will be examined in the T cell depleted thymus and an animal model for human oral tolerance. The proposed studies represent a unified approach to examine the function of these pathways in T cell immune regulation.//
